Job Description:

Our mission at Ansa Biotechnologies is to revolutionize the way we make DNA. Compared to reading or editing life's code, writing new code from scratch has remained one of the field's most elusive challenges. At Ansa, we are putting nature to work, creating novel enzymatic solutions that will redefine how we build DNA. We believe that the biggest challenges our world faces will be solved using biotechnology and that DNA synthesis is the fuel for that transformation. Our enzyme-based approach (published in Nature Biotechnology) promises to dramatically accelerate innovation in biological research and biotechnology, including therapeutics, diagnostics, biomanufacturing, and synthetic biology.

Developing and executing robust protein production processes will be instrumental in providing the key reagents needed to implement the fastest, cleanest, and most accurate DNA synthesis system in the world. As a Scientist in the Protein Production team, you will have the exciting opportunity to transition our protein production from research to pilot scale and beyond. You will make a critical impact on this transition, have the exciting opportunity to take part in a transition from research to support the work, and establish tools, systems, protocols to increase production of our key reagents.

Responsibilities

  • Operate equipment for, and implement scale-up of lysis, homogenization, clarification, chromatographic purification, and final buffer exchange to produce gram-scale batches of high quality, pure protein
  • Innovate on currently implemented downstream processes to drive down costs, decrease time of production, and improve purity and yield
  • Use experienced judgment to guide decision making regarding pipeline development as it relates to cost-benefit analysis, operational and technical capabilities
  • Help guide junior associates in downstream processing activities
  • Collaborating in a team environment: At Ansa, communication between teams is critical–our technology is built on enzymes, chemistry, and automation. Additionally, we believe that kind-natured honesty and openness are an instrumental part of the scientific endeavor.

Experience

  • PhD in Biochemistry, Microbiology, Bioengineering, Chemical Engineering or equivalent or Bachelor's/Master's with 5-10+ years of industry experience
  • 2+ years experience in downstream processing of proteins out of microbes
  • Experience in recombinant protein production
  • Experience with lysis using high shear techniques (eg. Microfluidizer), clarification (depth filtration or continuous flow centrifugation), column packing (eg. AxiChrom, BPG, INdEX), protein purification (AKTA FPLC, various chromatographic steps), and buffer exchange (TFF) at gram-level scale, with sub-kilogram level scale preferred
  • Experience with microbial expression systems
  • Experience managing 100+L scale buffer preparation is a plus
  • Experience with quality control assays is a plus
  • Experience with high density microbial fermentation is a plus
  • Experience with DNA-binding proteins is a plus
  • Enthusiasm to learn new techniques and curiosity to explore new areas of biology
